While pushing my bike around in neutral today, I noticed a sound coming from my bike.  I put it up on the centerstand, with the engine off, and spun the rear wheel.  I get repeated 'tok' sound coming from somewhere, I think the front sprocket.  I adjusted chain tension a few days ago, and I think the chain might be a tight now, only has a cm or so of play.  Could this be the cause?  If I need to readjust the chain tension, it's easy to tighten the tension, but how do I loosen it? Do I need to loosen the rear axle nut?

sounds like a dry link in your chain clean and lube the chain and yes you will have to loosen rear axle
