Dynojet needles...
They have a clip mounted to the needle with grooves in the needle head. Dynojet does not explain anywhere what effect moving the needle has. Does moving the needle have the same effect as placing washers under the needle?
I want to richen my mix (I know the smix screws won't help much (they only effect idle?) so what would be the best route to take. I've got the largest sized main jets dynojet has in the jet kit, and I've got 40 pilots on the way right now... should I shim the needle or move the clip up or down?

There's some pretty good threads here on jetting. do a search and there is a comprehensive list of jet sizes depending on your setup.
Not sure why u are using the biggest mains. do you have the pod filters?
To richen the mixture (1/4 to 3/4 throttle) you could lower the clip (thus raising the needle). I would think that moving the e-clip is the same as adding shims.
Check out factory pros site on how to tune carbs. This might help you.http://www.factorypro.com/tech/carbtun.html Basically you need to get the right size main jet before tuning the needle and so-forth. Biggest is not necessarily best.
The mixture screw basically affects idle.

Yep. To richen the mixture at mid-throttle, lower the e-clip. See this page for a detailed explanation:
